Giles-Harris began his football career at Duke University, where he played college football for the Duke Blue Devils. He made a name for himself as a standout linebacker, earning various accolades and recognition for his performances on the field. During his time at Duke, he was a two-time First Team All-ACC selection and was named to the All-ACC Academic Team.
